    Establishment of Micropropagation System for Mulberry (Morus alba L.)

    Mulberry (Morus alba L.) is an economically and ecologically significant woody perennial widely cultivated for sericulture, pharmaceuticals, and nutraceutical applications. However, conventional propagation methods are limited by low multiplication rates, genetic heterogeneity, and vulnerability to pests and diseases. This study aimed to develop a standardised and efficient micropropagation protocol for M. alba to support large-scale, uniform plant production. Explants (nodes, stems, and leaves) from healthy threeyear- old mother plants were subjected to ten surface sterilisation treatments using combinations of Clorox (10%), AgNO₃ (0.1 g/L), Tween 20, fungicides, and antibiotics. The optimised sterilisation method achieved a contamination rate of <5% and a survival rate >90%. Murashige and Skoog (MS) medium supplemented with 5.0 mg/L 6-Benzylaminopurine (BAP) and 0.2 mg/L Naphthalene Acetic Acid (NAA) yielded the highest shoot induction and shoot biomass across nodal and leaf explants. Significant callus formation was observed in nodal and stem explants, while browning -particularly in leaf cultures was mitigated using Woody Plant Medium and citric acid.

    An Improved Protocol for Callus Initi ati on Using Leaf Discs and Stems as Explants from Yellow Passion Fruit (Passifl ora edulis f. fl avicarpa)

    Passifl ora edulis f. fl avicarpa is a perennial vine mainly grown in tropical countries which has agricultural and horti cultural importance. Yet, the unavailability of an effi cient micropropagati on technique is a major obstacle to overcome. Diff erent surface sterilizati on methods were tested for the stem and leaf explants. To determine the best plant growth regulator concentrati ons for callus inducti on, diff erent combinati ons of NAA (2 to 5 mg/L) and BAP (0.1 to 0.3 mg/L) were tested. Murashige and Skoog full-strength medium was used as the basal medium for cultures. The use of 10% Clorox™ at 150 rpm for 12 minutes twice, followed by 0.1 g/L AgNO3 for 12 minutes found to be the most eff ecti ve surface sterilizati on method for leaves with a 100% survival rate. Results found 0.2 mg/L NAA; 2 mg/L BAP and 0.1 mg/L NAA; 4 mg/L BAP to be opti mal for callus inducti on from leaves under light and dark conditi ons with calli inducti on rates of 33.33% and 16.67%, respecti vely. For stems best results were obtained for 0.3 mg/L NAA; and 2 mg/L BAP with a 66.67% calli inducti on rate in light conditi ons and 33.33% in dark conditi ons. Calli were obtained from both explants within an average of 38.5 days and 44 days in light and dark conditi ons respecti vely. Calli of yellow passion fruit can be obtained using full MS media supplemented with NAA ranging from 0.1 to 0.3 mg/L and BAP ranging from 2 to 4 mg/L. Despite successes, vitrifi cati on and contaminati on underscore the need for refi nements on this protocol.

    An Efficient Plant Regeneration Using Leaf Explant for Osbeckia octandra (L.) DC; A Valuable Medicinal and Ornamental Plant

    Osbeckia octandra (Heen Bovitiya), which is one of the most valuable ayurvedic medicinal and ornamental plants in Sri Lanka is now threatened due to overexploitation in its natural habitat. In order to preserve this economically important plant species, a highly efficient in vitro clonal propagation system using leaf explants was developed in this study. The effects of three different concentrations of plant growth regulators: (6-benzylaminopurine (BAP), 1-naphthaleneacetic acid (NAA) and kinetin) and incubation conditions on plant regeneration were investigated. The plant growth parameters were analyzed in 15 biological replicates using one- way or two-way ANOVA. The present study revealed that the highest number of shoots per leaf explant with 92.4% shoot induction rate was achieved when young mature leaf explants were inoculated on Murashige and Skoog (MS) medium supplemented with 3 mg/L BAP and 0.5 mg/L NAA under dark conditions. When micro shoots were separated and subcultured on hormone-free MS medium with high sugar content (4%), shoots multiplied rapidly and a vigorous root system was developed under light conditions. The in vitro grown plantlets were successfully acclimatized and 89% of the regenerated plantlets survived. Our novel clonal propagation system will open new avenues for large- scale multiplication and improvement of medicinal and ornamental characteristics of this valuable species using biotechnological tools.
